DAP, also known as Death-associated protein 1 (DAP-1), plays a crucial role in ribosome hibernation, a process that stabilizes ribosomes in an inactive state, preventing their degradation. This occurs through its interaction with eiF5a (EIF5A and EIF5A2) at the ribosome's polypeptide exit tunnel, hindering mRNA translation. DAP is involved in ribosome hibernation during oocyte maturation, leading to ribosome inactivation and halting translation. This is vital as oocytes store large quantities of ribosomes produced during oogenesis for later use in early embryonic development. Additionally, DAP acts as a negative regulator of autophagy and mediates interferon-gamma-induced cell death.
