CYTH4
Description
The CYTH4 (cytohesin 4) is a protein-coding gene located on chromosome 22.
CYTH4 is a protein encoded by the CYTH4 gene in humans. It belongs to the cytohesin (CYTH) family, previously known as the PSCD (pleckstrin homology, Sec7 and coiled-coil domains) family. CYTH family members share a similar structure with an N-terminal coiled-coil motif, a central Sec7 domain, and a C-terminal pleckstrin homology (PH) domain. The coiled-coil motif facilitates homodimerization, the Sec7 domain exhibits guanine-nucleotide exchange protein (GEP) activity, and the PH domain interacts with phospholipids, anchoring CYTHs to membranes. These proteins are involved in regulating protein sorting and membrane trafficking. CYTH4 displays GEP activity in vitro with both ARF1 and ARF5 but not with ARF6. The gene structures of CYTH4 and CYTH1 are very similar.
CYTH4 promotes guanine-nucleotide exchange on ARF1 and ARF5, activating ARF factors by replacing GDP with GTP.
