PLN, also known as phospholamban, plays a critical role in regulating calcium levels within the heart muscle cells. It reversibly inhibits the activity of ATP2A2, a calcium pump located in the sarcoplasmic reticulum, by reducing ATP2A2's affinity for calcium ions. This inhibition is influenced by PLN's oligomeric state, with phosphorylation of PLN alleviating the inhibition. PLN's modulation of ATP2A2 activity impacts the contractility of heart muscle in response to physiological stimuli and regulates calcium re-uptake during muscle relaxation, contributing to calcium homeostasis within the heart muscle. Additionally, PLN influences intracellular calcium levels in elongated spermatids, suggesting a potential role in germ cell differentiation.
