CDK4 mutations are rare and contribute to a small percentage of melanoma cases.The CDK4 gene, located on chromosome 12q14.1, encodes the cyclin-dependent kinase 4 (CDK4) protein, which plays a key role in regulating the cell cycle progression through the G1 phase. CDK4 forms complexes with cyclin D proteins, allowing it to phosphorylate and inactivate the retinoblastoma protein (RB), thus promoting cell cycle progression into the S phase.
