It belongs to the ARF gene family, which encodes small guanine nucleotide-binding proteins involved in vesicular trafficking and activating phospholipase D. ARF3 is categorized as a class I ARF protein, along with ARF1 and ARF2. The ARF3 gene contains five exons and four introns. ARF3 has been shown to interact with other proteins.

ARF3 is a GTP-binding protein that activates the catalytic subunit of cholera toxin, an ADP-ribosyltransferase. It plays a role in protein trafficking, potentially regulating vesicle budding and uncoating within the Golgi apparatus.
