PPP2CA
Description
The PPP2CA (protein phosphatase 2 catalytic subunit alpha) is a protein-coding gene located on chromosome 5.
The PPP2CA gene encodes the catalytic subunit alpha of protein phosphatase 2A (PP2A). PP2A is a major Ser/Thr phosphatase involved in regulating cell growth and division. It consists of a core enzyme composed of a catalytic subunit and a regulatory subunit that can associate with various regulatory subunits.
PP2A-alpha is the major phosphatase for microtubule-associated proteins (MAPs). It can regulate the activity of various kinases, including phosphorylase B kinase, casein kinase 2, mitogen-stimulated S6 kinase, and MAP-2 kinase. It cooperates with SGO2 to protect centromeric cohesin during meiosis I. PP2A-alpha can dephosphorylate proteins like SV40 large T antigen and p53/TP53. It activates RAF1 by dephosphorylating it at 'Ser-259'. It also mediates dephosphorylation of WEE1, preventing its degradation and promoting the G2/M checkpoint. PP2A-alpha can dephosphorylate MYC, promoting its degradation, and FOXO3, promoting its stabilization. It catalyzes dephosphorylation of the pyrin domain of NLRP3, promoting NLRP3 inflammasome assembly.
