Description
Agap1 (ArfGAP with GTPase-activating protein and pleckstrin homology domains, 1) is a protein-encoding gene located on chromosome 11q13. It plays a crucial role in regulating the activity of small GTPases involved in signal transduction pathways. These pathways control numerous cellular processes, including cell growth, differentiation, and survival.
Agap1 specifically binds to and activates the GTPase-activating protein (GAP) activity of Arf6, a small GTPase involved in membrane trafficking and cytoskeletal dynamics. By activating Arf6-GAP, Agap1 promotes the conversion of Arf6-GTP to Arf6-GDP, leading to the inhibition of Arf6 signaling.

Mutations in the Agap1 gene have been linked to several neurological disorders, including:
- Intellectual disability: Agap1 mutations are responsible for approximately 1% of cases of non-syndromic intellectual disability, a condition characterized by significant impairments in cognitive and adaptive functioning.
- Autism spectrum disorder (ASD): Agap1 mutations have been associated with an increased risk of ASD, a developmental disorder characterized by social, communication, and behavioral challenges.
- Rett syndrome: Mutations in Agap1 have been identified in a subset of individuals with Rett syndrome, a severe neurodevelopmental disorder primarily affecting girls.
- Schizophrenia: Agap1 is considered a candidate gene for schizophrenia, a chronic mental disorder marked by hallucinations, delusions, and disorganized thinking.
- Epilepsy: Agap1 mutations have been linked to increased susceptibility to epilepsy, a neurological disorder characterized by recurrent seizures.
