Description

The FBXO11 (F-box protein 11) is a protein-coding gene located on chromosome 2.

FBXO11 is a protein encoded by the FBXO11 gene in humans. It belongs to the F-box protein family, which are components of the SCF (SKP1-cullin-F-box) ubiquitin ligase complex. FBXO11 mediates the degradation of substrate proteins, including BCL6, CDT2, and Snail, which are involved in cell cycle regulation, tumorigenesis, and metastasis. Inactivation of FBXO11-mediated BCL6 degradation contributes to abnormal germinal-center formation and tumorigenesis.

FBXO11 is a component of the SCF (SKP1-CUL1-F-box protein) E3 ubiquitin-protein ligase complex, which tags proteins for degradation by the proteasome. It targets a variety of proteins, including DTL/CDT2, BCL6, and PRDM1/BLIMP1. The SCF(FBXO11) complex plays a role in the terminal differentiation of germinal center B-cells into memory B-cells and plasma cells by degrading BCL6. It also regulates TGF-beta signaling, cell migration, and cell cycle progression by degrading DTL. FBXO11 further interacts with p53, inhibiting its transcriptional activity. FBXO11 is involved in erythropoiesis by degrading BAHD1, a heterochromatin-associated protein, and regulates macrophage cell death and inflammation in response to bacterial toxins. It also controls MHC-II levels by degrading CIITA, and participates in the antiviral response by activating the TBK1-IRF3-IFN-I axis.
