Description

The EIF4EBP3 (eukaryotic translation initiation factor 4E binding protein 3) is a protein-coding gene located on chromosome 5.

EIF4EBP3 is a protein encoded by the EIF4EBP3 gene in humans. It belongs to the EIF4EBP family, known for binding to eukaryotic initiation factor 4E (EIF4E) and inhibiting its assembly into the EIF4F complex. This gene is transcribed alongside its neighboring upstream gene (MASK), producing a transcript (MASK-BP3) that encodes a fusion protein with the MASK protein sequence and a modified C-terminus due to a different reading frame for the EIF4EBP3 segments.

EIF4EBP3 acts as a repressor of translation initiation by controlling the activity of EIF4E. Its hypophosphorylated form binds strongly to EIF4E, preventing the formation of the eIF4F complex and thus halting translation. Conversely, hyperphosphorylation causes EIF4EBP3 to detach from EIF4E, enabling the interaction of EIF4G1/EIF4G3 with EIF4E, initiating translation. Furthermore, EIF4EBP3 inhibits the export of mRNA from the nucleus mediated by EIF4E.
