Description

The DPEP3 (dipeptidase 3) is a protein-coding gene located on chromosome 16.

Dipeptidase 3 (DPEP3) is a protein encoded by the DPEP3 gene in humans. It's a membrane-bound glycoprotein belonging to the dipeptidase family (EC 3.4.13.19). DPEP3 plays a role in the breakdown of various dipeptides, including antibiotics like penem and carbapenem. This gene resides on chromosome 16, grouped with another member of the family, DPEP2. Multiple transcript variants, encoding different isoforms, have been identified for this gene.

DPEP3 lacks dipeptidase activity, unable to break down cystinyl-bis-glycine, leukotriene D4, or the antibiotic imipenem. This lack of activity is likely due to two key amino acid substitutions. Asparagine at position 359, instead of aspartate found in DPEP1/2, prevents it from acting as an acid/base catalyst, hindering water/hydroxide activation. Additionally, tyrosine at position 269 (instead of histidine) weakens the bond with zinc, possibly causing steric hindrance for substrate binding.

DPEP3 is also known as MBD3.
