The DGKI (diacylglycerol kinase iota) is a protein-coding gene located on chromosome 7.

DGKI, also known as diacylglycerol kinase iota or DGK-iota, is a protein that converts diacylglycerol (DAG) into phosphatidic acid (PA). It regulates the levels of these two bioactive lipids, which act as second messengers in cellular signaling pathways. DGKI has no preference for specific diacylglycerols and plays a key role in switching between different signaling pathways, often with opposing effects. By controlling the activation of RASGRP3, DGKI negatively regulates the Rap1 signaling pathway. It may also play a role in presynaptic DAG signaling and control neurotransmitter release during metabotropic glutamate receptor-dependent long-term depression.

DGKI is also known as DGK-IOTA.
