Description

The DENND3 (DENN domain containing 3) is a protein-coding gene located on chromosome 8.

DENND3 is a guanine nucleotide exchange factor (GEF) that activates RAB12 by promoting the exchange of GDP to GTP, converting inactive GDP-bound RAB12 into its active GTP-bound form. It plays a role in regulating autophagy in response to starvation by activating RAB12 through ULK1/2-dependent phosphorylation of Ser-472 and Ser-490, which allows the recruitment of 14-3-3 adapter proteins and upregulates GEF activity. DENND3 also participates in protein transport from recycling endosomes to lysosomes, regulating the degradation of the transferrin receptor and the amino acid transporter PAT4. Starvation-induced phosphorylation at Tyr-858 further enhances GEF activity and initiates autophagy. Evidence for these functions comes from UniProtKB:A2RT67 and PubMed:20937701.
