DDI1, or DNA-damage inducible 1 homolog 1 (S. cerevisiae), is a protein encoded by the DDI1 gene in humans.

DDI1 is a probable aspartic protease that acts as a proteasomal shuttle, linking the proteasome to replication fork proteins such as RTF2. It is essential for cell survival after replication stress, working in concert with DDI2 to remove RTF2 from stalled forks, allowing cell cycle progression and preserving genomic integrity.
