Description
The CSNK1E (casein kinase 1 epsilon) is a protein-coding gene located on chromosome 22.
Casein kinase I isoform epsilon is an enzyme encoded by the CSNK1E gene in humans. It is a serine/threonine protein kinase belonging to the casein kinase I protein family. This family is involved in regulating cytoplasmic and nuclear processes like DNA replication and repair. The enzyme is found in the cytoplasm as a monomer and can phosphorylate various proteins, including itself. It has been shown to phosphorylate proteins in the Period family, which are crucial for circadian rhythms. A homologous protein in Drosophila melanogaster, known as doubletime, also plays a role in circadian rhythm protein phosphorylation. Two transcript variants encoding the same protein have been identified for this gene. CSNK1E has been shown to interact with PER1 and AXIN1. Selective inhibitors of CSNK1E include PF-4800567, while non-selective inhibitors include PF-670462, which also inhibits CK1-δ.
Casein kinases are defined by their preference for acidic proteins like caseins as substrates. This protein is involved in Wnt signaling, phosphorylating DVL1, DVL2, and NEDD9/HEF1. It plays a key role in the circadian clock, balancing with PP1 to regulate the phosphorylation of PER1 and PER2, thereby controlling their nuclear transport and degradation. Additionally, it inhibits cytokine-induced granulocytic differentiation.
