Description

The CREB3L2 (cAMP responsive element binding protein 3 like 2) is a protein-coding gene located on chromosome 7.

CREB3L2 is a transcription factor involved in the unfolded protein response (UPR). In the absence of endoplasmic reticulum (ER) stress, it is inserted into ER membranes with its N-terminal DNA-binding and transcription activation domains facing the cytoplasm. When ER stress occurs, CREB3L2 is transported to the Golgi apparatus, where it is cleaved by proteases S1P/MBTPS1 and S2P/MBTPS2. The released N-terminal fragment then translocates to the nucleus and activates the transcription of specific target genes. CREB3L2 plays a crucial role in chondrogenesis by activating SEC23A transcription, promoting the transport and secretion of cartilage matrix proteins. It also potentially regulates ER biogenesis-related genes. In a neuroblastoma cell line, CREB3L2 protects cells from ER stress-induced death. In vitro, it directly binds to the CRE site and activates the transcription of target genes.
