COPS7B is a component of the COP9 signalosome complex (CSN), which plays a crucial role in regulating various cellular and developmental processes. CSN acts as a key regulator of the ubiquitin (Ubl) conjugation pathway by mediating the removal of NEDD8 from cullin subunits of SCF-type E3 ligase complexes. This de-neddylation process leads to a decrease in the ubiquitin ligase activity of SCF-type complexes, such as SCF, CSA, and DDB2. The CSN complex is also involved in the phosphorylation of several proteins, including p53/TP53, JUN, I-kappa-B-alpha/NFKBIA, ITPK1, and IRF8/ICSBP, potentially through its interaction with CK2 and PKD kinases. CSN-dependent phosphorylation of TP53 promotes its degradation by the Ubl system, while phosphorylation of JUN protects it from degradation.
