The CNOT7 (CCR4-NOT transcription complex subunit 7) is a protein-coding gene located on chromosome 8.
CCR4-NOT transcription complex subunit 7 is a protein that in humans is encoded by the CNOT7 gene. It is a subunit of the CCR4-Not deadenylase complex. The protein encoded by this gene binds to an anti-proliferative protein, B-cell translocation protein 1, which negatively regulates cell proliferation. Binding of the two proteins, which is driven by phosphorylation of the anti-proliferative protein, causes signaling events in cell division that lead to changes in cell proliferation associated with cell-cell contact. The protein has both mouse and yeast orthologs. Alternate splicing of this gene results in two transcript variants encoding different isoforms. CNOT7 has been shown to interact with: BTG1, PABPC1, TOB1, and TOB2.
CNOT7 is a catalytic component of the CCR4-NOT complex, a major cellular mRNA deadenylase involved in various processes such as bulk mRNA degradation, miRNA-mediated repression, translational repression during initiation, and general transcription regulation. CNOT7 exhibits 3'-5' poly(A) exoribonuclease activity for synthetic poly(A) RNA substrates. Its function is partially redundant with CNOT8. The CCR4-NOT complex acts as a translational repressor during miRNA-mediated repression and initiation. CNOT7's influence on mRNA expression contributes to additional complex functions. CNOT7 associates with BTG family members, particularly TOB1 and BTG2, and is essential for their anti-proliferative activity.
