The CNOT6L (CCR4-NOT transcription complex subunit 6 like) is a protein-coding gene located on chromosome 4.

CCR4-NOT transcription complex subunit 6 like is a protein that in humans is encoded by the CNOT6L gene. It is a paralog of CNOT6 and therefore a potential subunit of the CCR4-Not deadenylase complex.

CNOT6L possesses 3'-5' poly(A) exoribonuclease activity towards synthetic poly(A) RNA substrates. It acts as a catalytic component of the CCR4-NOT complex, a key cellular mRNA deadenylase implicated in various cellular processes, including bulk mRNA degradation, miRNA-mediated repression, translational repression during translational initiation, and general transcription regulation. Its influence on mRNA expression contributes to additional complex functions. CNOT6L may participate in the deadenylation-dependent degradation of mRNAs via the 3'-UTR AU-rich element-mediated mechanism and is involved in the deadenylation-dependent degradation of CDKN1B mRNA. Its mRNA deadenylase activity can be inhibited by TOB1. CNOT6L plays a role in cell proliferation, cell survival, and prevention of cellular senescence.
