The CNOT6 (CCR4-NOT transcription complex subunit 6) is a protein-coding gene located on chromosome 5.

CNOT6 is a human gene encoding CCR4-NOT transcription complex subunit 6. This protein is a subunit of the CCR4-Not complex involved in transcriptional regulation and deadenylation. It exhibits 3'-5' exonuclease activity, preferring polyadenylated substrates.

CNOT6 is a poly(A) nuclease with 3'-5' RNase activity. As a catalytic component of the CCR4-NOT complex, it plays a crucial role in mRNA degradation, miRNA-mediated repression, translational repression during initiation, and general transcription regulation. Its influence on mRNA expression contributes to diverse cellular processes. CNOT6 is involved in mRNA decay mediated by the mCRD of the FOS gene in the cytoplasm. In the presence of ZNF335, it enhances the ligand-dependent transcriptional activity of nuclear hormone receptors, such as RARA. While the increase in ligand-dependent ESR1-mediated transcription is less significant, CNOT6 contributes to cell proliferation, survival, and prevents cellular senescence.
