The CNOT1 (CCR4-NOT transcription complex subunit 1) is a protein-coding gene located on chromosome 16.

CCR4-NOT transcription complex subunit 1 is a protein that in humans is encoded by the CNOT1 gene. It is a part of the CCR4-Not complex, which deadenylates mRNAs. CNOT1 acts as a scaffold protein, binding other subunits of the complex. CNOT1 has been shown to interact with CNOT8.

CNOT1 is a scaffolding protein that is part of the CCR4-NOT complex, a major cellular mRNA deadenylase involved in various cellular processes. These processes include bulk mRNA degradation, miRNA-mediated repression, translational repression during translational initiation, and general transcription regulation. Its scaffolding function facilitates the interaction of the CCR4-NOT complex with diverse RNA-binding proteins, targeting the complex to specific mRNA 3'UTRs. CNOT1 plays a role in the degradation of AU-rich element (ARE)-containing mRNAs, likely through its association with ZFP36. It mediates the recruitment of the CCR4-NOT complex to miRNA targets and the RISC complex via interactions with TNRC6A, TNRC6B, or TNRC6C. Acting as a transcriptional repressor, CNOT1 suppresses ligand-dependent transcriptional activation by nuclear receptors. Furthermore, CNOT1 contributes to the maintenance of embryonic stem (ES) cell identity.
