Description

The CLEC12B (C-type lectin domain family 12 member B) is a protein-coding gene located on chromosome 12.

CLEC12B acts as an inhibitory receptor, negatively regulating both immune and non-immune processes. Upon phosphorylation, it recruits PTPN6 and PTPN11 phosphatases through its ITIM motif, counteracting activation signals. While it inhibits KLRK1/NKG2D signaling, it doesn't bind known ligands of KLRK1/NKG2D, indicating it's not its specific inhibitor. CLEC12B may limit myeloid cell subset activation during infection or inflammation, potentially protecting cells from natural killer cell lysis. Furthermore, it may regulate melanocyte cell cycle and differentiation by inactivating STAT3.
