The CFLAR (CASP8 and FADD like apoptosis regulator) is a protein-coding gene located on chromosome 2.

CFLAR, also known as c-FLIP, is a protein encoded by the CFLAR gene in humans. It functions as a regulator of apoptosis, playing a critical role in the balance between cell survival and death.

CFLAR, also known as c-FLIP, is an apoptosis regulator protein that plays a crucial role in mediating the balance between cell survival and death pathways in mammalian cells. It acts as an inhibitor of apoptosis triggered by TNFRSF6, preventing the activation of caspase-8. A proteolytic fragment of CFLAR (p43) remains associated with the death-inducing signaling complex (DISC) to prevent further recruitment and processing of caspase-8. Interestingly, both full-length and shorter isoforms of CFLAR can either induce apoptosis or reduce TNFRSF-triggered apoptosis, highlighting its complex regulatory role. Notably, CFLAR lacks enzymatic (caspase) activity.
