Ceramide synthase 6 is a protein that in humans is encoded by the CERS6 gene.
Ceramide synthase 6 (CerS6) is an enzyme that catalyzes the transfer of a fatty acid chain from acyl-CoA to a sphingoid base, with a strong preference for palmitoyl-CoA (C16:0-CoA). It can utilize other acyl donors, but with lower efficiency. CerS6 is involved in both the de novo synthesis and salvage pathways of ceramide biosynthesis, producing dihydroceramides and ceramides, respectively. Ceramides generated by CerS6 are implicated in inflammation, metabolic regulation, and hepatic lipid accumulation. Under high-fat diets, CerS6-produced palmitoyl-ceramides bind to the mitochondrial fission factor (MFF), triggering mitochondrial fragmentation and contributing to obesity development.
