CDK5RAP3
Description
The CDK5RAP3 (CDK5 regulatory subunit associated protein 3) is a protein-coding gene located on chromosome 17.
CDK5 regulatory subunit-associated protein 3 is a protein that in humans is encoded by the CDK5RAP3 gene. Neuronal CDC2-like kinase, which is involved in the regulation of neuronal differentiation, is composed of a catalytic subunit, CDK5, and an activating subunit, p25NCK5A. The protein encoded by this gene binds to p25NCK5A and therefore may be involved in neuronal differentiation. The encoded protein, which may be a substrate of neuronal CDC2-like kinase, has also been found in vascular endothelial cells, where it mediates cell proliferation.
CDK5RAP3 is a substrate adapter for ufmylation, the covalent attachment of the ubiquitin-like modifier UFM1 to substrate proteins, in response to endoplasmic reticulum stress. It negatively regulates NF-kappa-B-mediated gene transcription through the control of RELA phosphorylation. CDK5RAP3 is a probable tumor suppressor initially identified as a CDK5R1 interactor controlling cell proliferation. It also regulates mitotic G2/M transition checkpoint and mitotic G2 DNA damage checkpoint. Through its interaction with CDKN2A/ARF and MDM2, CDK5RAP3 may induce MDM2-dependent p53/TP53 ubiquitination, stabilization and activation in the nucleus, thereby promoting G1 cell cycle arrest and inhibition of cell proliferation. It may also play a role in the rupture of the nuclear envelope during apoptosis. CDK5RAP3 may regulate MAPK14 activity by regulating its dephosphorylation by PPM1D/WIP1. It is required for liver development.
