The CDC42BPA (CDC42 binding protein kinase alpha) is a protein-coding gene located on chromosome 1.

CDC42BPA gene encodes Serine/threonine-protein kinase MRCK alpha which is a member of the Serine/Threonine protein kinase family. This kinase contains multiple functional domains, including a kinase domain similar to the myotonic dystrophy protein kinase (DMPK), a Rac interactive binding (CRIB) domain, and binds to CDC42. It may function as a CDC42 downstream effector mediating CDC42 induced peripheral actin formation, and promoting cytoskeletal reorganization.

CDC42BPA, also known as MRCK alpha, is a serine/threonine protein kinase that plays a crucial role in regulating cytoskeleton reorganization and cell migration. It acts as a downstream effector of CDC42, a small GTPase involved in various cellular processes. CDC42BPA regulates actin cytoskeletal reorganization by phosphorylating specific proteins, including PPP1R12C and MYL9/MLC2. In conjunction with MYO18A and LURAP1, it modulates lamellar actomyosin retrograde flow, which is essential for cell protrusion and migration. Additionally, CDC42BPA phosphorylates PPP1R12A, LIMK1, and LIMK2, and may contribute to TFRC-mediated iron uptake. It forms a complex with FAM89B/LRAP25, targeting LIMK1 to the lamellipodium, leading to its activation and subsequent phosphorylation of CFL1, which is critical for lamellipodial F-actin regulation. Furthermore, CDC42BPA triggers the formation of an apical actin ring during epithelial extrusion of apoptotic cells.
