CASP8AP2
Description
The CASP8AP2 (caspase 8 associated protein 2) is a protein-coding gene located on chromosome 6.
CASP8-associated protein 2 is a protein that, in humans, is encoded by the CASP8AP2 gene. This protein is highly similar to FLASH, a mouse apoptotic protein identified by its interaction with the death-effector domain (DED) of caspase 8. Research on FLASH protein suggested that this protein may be a component of the death-inducing signaling complex, which includes Fas receptor, Fas-binding adapter FADD, and caspase 8, and plays a regulatory role in Fas-mediated apoptosis. CASP8AP2 has been shown to interact with TRAF2.
CASP8AP2 plays a crucial role in various cellular processes, including apoptosis, inflammation, and cell cycle regulation. It participates in TNF-alpha-induced suppression of glucocorticoid receptor (GR) transactivation by inhibiting the activity of nuclear receptor coactivators NCOA2 and NCOA3. This protein also contributes to TNF-alpha-induced activation of NF-kappa-B through a TRAF2-dependent pathway. Furthermore, CASP8AP2 acts as a downstream mediator in CASP8-induced NF-kappa-B activation and is essential for the activation of CASP8 in FAS-mediated apoptosis. Notably, it is required for histone gene transcription and the progression through the S phase of the cell cycle.
