CARD16


Description

The CARD16 (caspase recruitment domain family member 16) is a protein-coding gene located on chromosome 11.

CARD16 is a caspase inhibitor that regulates the activation of procaspase-1 (CASP1). It plays a role in controlling the maturation and release of pro-interleukin-1 beta (IL1B) during inflammation. CARD16 inhibits the release of IL1B in response to lipopolysaccharide (LPS) in monocytes. It also activates NF-kappa-B, a key transcription factor involved in the pro-inflammatory cytokine response. CARD16 can inhibit CASP1-mediated neuronal cell death, as well as cell death triggered by TNF-alpha, hypoxia, UV radiation, and staurosporine. However, it does not prevent ER stress-mediated cell death. CARD16 achieves its inhibitory effects by preventing the activation of caspases CASP1 and CASP4, possibly by blocking the interaction between CASP1 and RIPK2.
