Description

The BDH2 (3-hydroxybutyrate dehydrogenase 2) is a protein-coding gene located on chromosome 4.

BDH2 is a NAD(H)-dependent dehydrogenase/reductase that prefers cyclic substrates. It catalyzes the conversion of 4-oxo-L-proline to cis-4-hydroxy-L-proline, a detoxification mechanism for ketoprolines. BDH2 also forms 2,5-dihydroxybenzoate (2,5-DHBA), a siderophore that binds to free cytoplasmic iron and interacts with LCN2, regulating iron transport, homeostasis, and protecting cells from oxidative stress. The iron-siderophore complex is transported into mitochondria, supplying iron for mitochondrial metabolic processes, including heme synthesis. BDH2 may also act as a 3-hydroxybutyrate dehydrogenase.
