The B3GALT5 (beta-1,3-galactosyltransferase 5) is a protein-coding gene located on chromosome 21.

B3GALT5 is a human gene that encodes the enzyme beta-1,3-galactosyltransferase 5. This enzyme belongs to the beta-1,3-galactosyltransferase (beta3GalT) gene family, which encodes type II membrane-bound glycoproteins. These proteins have diverse enzymatic functions using various donor substrates and acceptor sugars. B3GALT5 is closely related to the Drosophila Brainiac gene and contains its protein coding sequence within a single exon. The beta3GalT proteins share conserved sequences not found in other related proteins. The carbohydrate chains synthesized by these enzymes are classified as type 1, while other enzymes synthesize type 2 chains. The ratio of type 1 to type 2 chains varies during embryonic development. B3GALT5 is one of the most likely candidates for synthesizing type 1 Lewis antigens, which are often elevated in gastrointestinal and pancreatic cancers. While the encoded protein is inactive with N-linked glycoproteins, it plays a role in mucin glycosylation.
