Description
The ATG14 (autophagy related 14) is a protein-coding gene located on chromosome 14.
ATG14 is essential for both basal and inducible autophagy. It plays a key role in the localization of the autophagy-specific PI3-kinase complex PI3KC3-C1, and is involved in autophagosome formation and MAP1LC3/LC3 conjugation to phosphatidylethanolamine. ATG14 promotes BECN1 translocation from the trans-Golgi network to autophagosomes, and enhances PIK3C3 activity in a BECN1-dependent manner. It is also essential for the autophagy-dependent phosphorylation of BECN1, stimulating its phosphorylation while suppressing PIK3C3 phosphorylation by AMPK. ATG14 binds to the STX17-SNAP29 binary t-SNARE complex on autophagosomes and primes it for VAMP8 interaction, promoting autophagosome-endolysosome fusion. It is also involved in modulating hepatic lipid metabolism.
