ARRDC1

Description

The ARRDC1 (arrestin domain containing 1) is a protein-coding gene located on chromosome 9.

ARRDC1 is a protein that functions as an adapter, bringing together ubiquitin-protein ligases with their specific substrates. This protein plays a role in the incorporation of SLC11A2 into extracellular vesicles through a ubiquitination-dependent mechanism. ARRDC1 also plays a role in the extracellular transport of proteins between cells through the release of microvesicles. It negatively regulates the NOTCH signaling pathway by participating in the ITCH-mediated ubiquitination and subsequent degradation of NOTCH1. ARRDC1 interacts with several proteins, including ITCH, ARRB1, ARRB2, TSG101, WWP2, SLC11A2, WWP1, NEDD4, and PDCD6IP.

ARRDC1 functions as a molecular adapter that brings together ubiquitin-protein ligases with their specific targets. This process is crucial for regulating protein activity and degradation. For example, ARRDC1 helps incorporate SLC11A2 into extracellular vesicles, which are tiny packages that cells release. This involves adding a tag called ubiquitin to SLC11A2. Additionally, ARRDC1 plays a role in the general transport of proteins between cells through the release of microvesicles. Furthermore, ARRDC1 regulates the NOTCH signaling pathway by participating in the degradation of NOTCH1, a key protein involved in cell development and communication. This process is mediated by the ubiquitin ligase ITCH.
