ARHGAP18, also known as MacGAP or Rho-type GTPase-activating protein 18, functions as a Rho GTPase-activating protein that inhibits Rho activity. This inhibition suppresses F-actin polymerization, which is crucial for maintaining cell shape, spreading, and migration. ARHGAP18 plays a critical role in regulating cortical actomyosin network formation, thereby influencing tissue tension and 3D tissue shape. It also acts downstream of YAP1, inhibiting actin polymerization and consequently reducing YAP1's nuclear localization.
