TP73


Description

The TP73 (tumor protein p73) is a protein-coding gene located on chromosome 1.

TP73 is a protein closely related to the tumor suppressor protein p53. Due to its structural similarity to p53, it is also considered a tumor suppressor. It plays a role in cell cycle regulation and induction of apoptosis. Like p53, TP73 exists in various isoforms, resulting from alternative splicing and an alternative promoter in its DNA sequence. TP73, also known as tumor protein 73, was the first identified homolog of the tumor suppressor gene, p53. Similar to p53, TP73 has multiple variants. It is expressed in distinct forms with differences at either the C- or N-terminus. Currently, six different C-terminus splicing variants have been identified in normal cells. The TP73 gene encodes a protein with significant sequence homology and functional similarity to the tumor suppressor p53.

TP73 participates in the apoptotic response to DNA damage. Isoforms containing the transactivation domain are pro-apoptotic, isoforms lacking the domain are anti-apoptotic and block the function of p53 and transactivating p73 isoforms. It may be a tumor suppressor protein. TP73 is an activator of FOXJ1 expression and is an essential factor for the positive regulation of lung ciliated cell differentiation.
