The MDH2 (malate dehydrogenase 2) is a protein-coding gene located on chromosome 7.
MDH2, also known as malate dehydrogenase 2, encodes an enzyme that catalyzes the reversible oxidation of malate to oxaloacetate using the NAD/NADH cofactor system in the citric acid cycle. This mitochondrial protein is crucial for the malate-aspartate shuttle, which coordinates metabolism between the cytosol and mitochondria. The protein exists as a dimer, suggesting a strong connection between protein stability and enzymatic activity. Each subunit has two distinct domains: the NAD-binding domain (with a Rossmann fold motif) and the substrate binding domain. The active site is located in a cleft between these domains. Crystallography reveals a dimer interface composed of interacting alpha-helices, forming a compact structure.
