CRADD


Description

The CRADD (CASP2 and RIPK1 domain containing adaptor with death domain) is a protein-coding gene located on chromosome 12.

CRADD, encoded by the CRADD gene, is a death domain-containing protein that induces cell apoptosis. It interacts with caspase 2/ICH1 through its CARD domain, recruiting it to the cell death signal transduction complex. This complex includes tumor necrosis factor receptor 1 (TNFR1A), RIPK1/RIP kinase, and other CARD domain-containing proteins. CRADD has been shown to interact with RIPK1 and Caspase 2.

CRADD acts as an adapter protein, linking PIDD1 and caspase CASP2 to form the PIDDosome complex. This complex activates CASP2, triggering apoptosis. CRADD also recruits CASP2 to the TNFR-1 signaling complex through its interactions with RIPK1 and TRADD, potentially playing a role in the tumor necrosis factor-mediated signaling pathway.

CRADD is also known as MRT34, RAIDD.
