The CLDN11 (claudin 11) is a protein-coding gene located on chromosome 3.

Claudin-11 is a protein encoded by the CLDN11 gene in humans. It belongs to the claudin family, which are vital components of tight junctions, essential for maintaining cell barriers. Claudin-11 is particularly important in the central nervous system (CNS), as it is a major component of myelin, the protective sheath around nerve fibers. It contributes to the normal function of the CNS, hearing, and spermatogenesis. Evidence suggests Claudin-11 controls the permeability of myelin sheaths, potentially playing a role in cell growth and migration during development. It has been implicated as a potential target for autoimmune responses in demyelinating diseases. Studies in mice lacking Claudin-11 show behavioral changes, including altered responses in open field tests and sound localization, coupled with changes in neurotransmitter levels in brain regions involved in emotions and hearing. This suggests that changes in myelin membrane properties, even without degeneration, could contribute to neuropsychiatric disorders in humans.

Claudin-11 is crucial for tightly sealing the gaps between cells, preventing substances from leaking between them. This process occurs independently of calcium ions and relies on cell-to-cell adhesion.
