The CDKN2B (cyclin dependent kinase inhibitor 2B) is a protein-coding gene located on chromosome 9.

CDKN2B, also known as multiple tumor suppressor 2 (MTS-2) or p15INK4b, is a human gene that encodes a protein with cell growth regulatory functions. It is located adjacent to the tumor suppressor gene CDKN2A, in a region frequently mutated or deleted in various cancers. The encoded protein acts as a cyclin-dependent kinase inhibitor, specifically targeting CDK4 and CDK6. By forming a complex with these kinases, it prevents their activation by cyclin D, thus inhibiting cell cycle progression at the G1 phase. Its expression is strongly induced by TGF beta, suggesting its role in TGF beta-mediated growth inhibition. Two alternatively spliced transcripts exist, resulting in proteins with identical N-terminal sequences but differing in their C-terminal regions.
